Topic: Studying DNA Repair Inside Living Cells: Looking at Things Too Small to See
One of the hallmarks of cancer cells is their genome instability; instead of a normal set of 23 pairs of chromosomes, they have an astonishing number of chromosomal alterations, some of which create gene fusions that accelerate the transition of cells toward cancer. These changes arise in cells that have lost their normal ability to repair chromosomal damage - breaks in the DNA chain. My lab is interested in defining in molecular detail how chromosome breaks are normally repaired and what happens when this repair goes awry; what happens to the DNA and how is it acted upon by DNA repair enzymes. We study repair events in living cells, even though a single DNA chain is impossible to see with even the most powerful light microscope. Nevertheless we have found ways to "look at" these events in great detail.
